10W-40 is the grade Yamaha lists for the YZ250F, YZ450F, WR250F and WR450F, and what most trail and enduro four strokes call for. It is also the right grade for most Japanese four stroke minis.
Everything here is a motorcycle 4T oil for a wet clutch and a shared gearbox, rated JASO MA or MA2. The MWMX 10W-40 is a full synthetic PAO and ester blend, rated API SP and JASO MA2, in 1L and 4L.

Yamaha YZ250F, YZ450F, WR250F and WR450F, Kawasaki KX250 and KX450, Suzuki RM-Z, most Honda and Yamaha trail bikes and the four stroke minis. Check the manual; where it lists 10W-40 or 10W-50, either is fine.
Full synthetic holds up longer and protects better when the engine gets hot, which every race bike does. Mineral is fine for a trail bike on short intervals. Every MWMX oil is full synthetic.
About a litre with a filter change on the current YZ450F and YZ250F, so a 1L bottle does one change and the 4L does four. Check the sight glass or dipstick after a warm up.
No. Car oils carry friction modifiers that make a wet clutch slip. Use a motorcycle 4T oil rated JASO MA or MA2.
